Ben Davies and Kieffer Moore will miss Wales' final World Cup qualifying group games against Liechtenstein and North Macedonia due to injury.
Swansea City defender Ben Cabango has also withdrawn.
Tottenham Hotspur defender Davies, 32, has not played since damaging his hamstring during October's defeat by Belgium.
He was included in Wales' squad for the trip to Liechtenstein on Saturday, 15 November, and the home match against North Macedonia the following Tuesday.
Wrexham striker Moore, 33, was also named in the squad but sustained an injury during his club's Championship win over Charlton Athletic last Saturday.
He has been replaced by uncapped Cardiff City forward Isaak Davies, while Queens Park Rangers left-back Rhys Norrington-Davies comes in for Spurs defender Davies.
Davies' withdrawal also means Wales will need to find a new captain as he has been leading the team in the continued absence of regular captain Aaron Ramsey through injury.
Swansea skipper Cabango played the full 90 minutes in Saturday's 4-1 defeat by Ipswich Town but the Football Associaition of Wales (FAW) say the 25-year-old also has an injury.

From being a stroll in the park...easy three points...the fact that if we win by six goals, (which will make Tuesday's game a lot more comfortable) will add a bit of zip to the game.
An early goal, followed by two more before half time, should set us up for a classic second half.
Wales seldom seem to do things the easy way...so we'll probably get five up with ten mins left, and then it'll be a nail biting last ten.
Either that, or Bellamy will just take the three points, and bring on a load of subs, protecting his "star' players for the big game...leaving us in a must win situation against draw specialists NM.
We might be a better team with Johnson or Brooks playing in the middle up front.
Play with the ball on the deck
I just don't see this team scoring like that, it's hard to find exact numbers but it looks like we have scored 6 or more barely more than a handful of times in our history with at least half coming in the 19th century.
I can see a dominant performance but a 2/3 nil.
Ultimately just make sure we win the game, there seems to have been a few instances over the last decade or so where we have scraped late wins in these sort of fixtures.

Feels like an ideal game for Colwill with Wilson out
From a quick websearch I think they do carry over into the playoffs but not the finals. It looks like Ben Davies, Rodon, Ampadu, Mepham and Jordan James are a yellow away from suspension. Big defensive impact is possible....
Am I right in believing the final two group games will almost certainly only determine whether Wales finish second or third in the table and we'll be in the play-offs either way?
Yes, this would make a massive difference, also then avoid the likes of Sweden and Serbia too I think. There is no way we put 6 on Liechtenstein , it would be wonderful if we did and make things a damn sight easier against North Macedonia but it’s just not what Wales historically do. Will be very tricky to break down North Macedonia in the 2nd game, I believe they are also guaranteed a play off spot if they finish 3rd but have just as much incentive to avoid the 3rd place finish as we do
